Toxicol.I have Examined the Effects of Treatment of Several Nucleotide Excision Repair (Ner) -Proficient (Ner+) and -Deficient (Ner-) Cell Lines with the Carcinogenic Metabolite (+) Anti-Dbpde. Repair Experiments were done Using the 32p-Postlabeling Assay. in Human Primary Fibroblasts, Treatment with (+) Anti-Dbpde Generated 1 Major (Approximately 65-70%) and at Least 3 Minor Dna Adducts That were Chromatographically Identical to Those Formed by Direct Treatment of 3’ Gmp and 3’ Amp with (+) Anti-Dbpde. in the Ner-Proficient Cells, Adduct Levels Declined Approximately 50% in 6 H After Carcinogen Removal; by 34 H, the Number of Adducts/109 Nucleotides was 20% of the Level at the Time of Carcinogen Removal. This Suggests That Persistent Dbp Adducts do not Block Normal Dna Replication. Our Results Also Suggest That the Extreme Carcinogenicity of Dbpde may Result from the Ability of Some Dbpde-Dna Adducts to Escape Repair And/Or be Bypassed by Dna Polymerases.
