• Developed Numerous Tcr-Gfp Transgenic Mouse Models for Adopted Transfer of Bone Marrow Into B6 Non-Irradiated Recipients to Generate Immune System Chimaeras (Developed Method) for Two Photon Imaging of T Cell Development Real Time in Live Intact Thymus. MY Studies were the First Visualization in Situ of Ongoing Thymopoiesis and Showed That Thymocytes Undergoing Positive Selection Migrate Through the Thymic Cortex by Way of 'Random Walk' Until Receiving Positive Selection Signals. Selection Signals Lead to Changes in Morphological and Migratory Behavior Resulting in the Directed Migration of Positively Selected Thymocytes Into the Medulla for Final Process of Negative Selection. This Behavior is Ccr7 Dependent. My Post-Doc Culminated in Commissioned Reviews on T Cell Development and on 3D Cell Tracking in Situ. Also, Commentary on the Importance of Quantifying Imaging Data and Use of Modeling in the Study of Biological Systems.
• Investigated the Role of the Master Regulator Protein Notch2 in Hematopoiesis. Using Both Over Expression Animal Models and Notch2+/- Animal Models i Showed the Critical Role Notch2 Plays in B1 B Cell Development, as Well its Aberrant Role in Tumorigenesis When Over-Expressed. i Employed A Wide Range of Expression Analysis Techniques to Determine Protein Expression Within the Immune System, Stem Cell Sorting and Retroviral Transduction of Stem Cell and Transplantation to Generate Notch2 Over-Expressing Animals. Also, Bone Marrow, Peripheral Blood and Organ Analysis Using Flow Cytometry.
