  • Activities and Societies: Teaching Fellow Mcdb 315b: Reaction to Injury, Mechanisms of Disease (Spring 2006) Head Teaching Fellow (Spring 2007, Spring 2008, Spring 2010)My Major Project Focused on Autosomal Dominant Polycystic Kidney Disease (Adpkd), the Most Prevalent Genetic Disease, for Which There are Currently no Fda-Approved Therapies. Renal Cyst Formation is Attributed to Both Excess Apical Fluid Secretion Into the Cyst Lumen and Inappropriate Proliferation of the Renal Epithelial Cells. These Processes are Thought to be Mediated by the Cftr Chloride Channel and the Mtor Pathway, Respectively. Both of These Targets Lie Downstream of and are Antagonized by Adenosine Monophosphate-Activated Protein Kinase (Ampk). WE Hypothesized That if Ampk is Stimulated, Then This will Further the Inhibition of Both Cftr and Mtor, Leading to Decreased Cystogenesis. Indeed, in Vitro, in Situ, and in Vivo, WE were Able to Demonstrate That Pharmacologic Stimulation of Ampk by Metformin, Results in Decreased Cyst Formation. These Studies Also have the Potential for Clinical Application as Metformin is Already Fda Approved for the Treatment of Other Conditions.
